Glycerol Bacteria Stocks

Brief Description

Long term storage of bacteria transformed with plasmid DNA.

Protocol Details

Reagents:

  • 80% glycerol (sterile)
  • (optional) liquid nitrogen or ethanol/dry ice

Basic Protocol

1. Grow 1-2 ml of culture overnight or to saturation in LB medium plus appropriate antibiotic.

2. Mix 850 µl of the culture with 200 µl of 80% glycerol. The rest may be used for plasmid preparation.

3. Freeze quickly in liquid nitrogen or ethanol/dry ice.

4. Store at -80°C.
